Automated 96-well protein quantification

Rapid, 96-well protein quantification has been optimized on the CyBio FeliX, facilitating high-throughput screening and identification of optimal solutions. This protein quantification is achieved using the Nano-Glo® HiBiT Extracellular Detection System from Promega. This assay permits precise quantification of extracellular and secreted HiBiT-tagged proteins.

Protein quantification on the CyBio FeliX

For high-throughput quantification, proteins of interest can be modified with the HiBiT tagging technology. HiBiT is a small, 11-amino-acid, epitope tag capable of producing a bioluminescent signal when bound to its complementation partner, LgBiT. 

Tagged proteins can then be quickly setup for quantification on the CyBio FeliX via the Nano-Glo® HiBiT Extracellular Detection System. For maximum throughput, cultures can be grown directly in 96-well plates and sampled directly with the 96 tip-tray format available with the CyBio FeliX. 

Standard curve generation

A standard curve by which the protein concentration of samples can be assessed is first generated. Reagents and the control protein are loaded onto the FeliX for the serial dilution.

Using a single tip, reagents are transferred to the plate, before a serial dilution is then performed.

The HiBit reagent buffers are then aliquoted by the CyBio FeliX into a 96-well format for parallel addition.

Quantifying more than one 96 well plate?

Simply load more plates onto the deck and quickly fill them all with 96 tip-tray instead of an 8-channel column. 

 

Sample quantification

Once the plate is pre-filled with HiBit Reagent, all that is remaining is to add the sample to be quantified. This can be purified protein or often cell culture supernatant.

With the 96 tip-tray, all your samples are transferred in parallel to the assay plate. This ensures any variation due to sample-reagent incubation time is minimized. 

Four separate 96 well plates can be processed without user intervention!
